i absolutely love medal of honor heroes 2 multiplayer. team deathmatch in the city. i've been playing my xbox 360 more now that i've gotten rock band 2 and guitar hero world tour, but i still love going back to the wii to play MOHH2. the point and shoot with the wiimote is the best....i have total control. i just wish it was easier to keep track of your friends, and that you could talk through a headset to your friends.

Well obvious choices are Mario Kart, Super Smash Bros, Goldeneye, Street Fighter, Bomberman, Worms, etc. But here are my two perhaps slightly uncommon suggestions for best multiplayer game:
Pac Man VS: One of the most underrated games of all-time in my opinion, my friends and I have had some of our most fun and frantic multiplayer sessions with this little gem. It's instantly accessible to anyone because everyone knows the basics of Pac-Man and because everyone desperately wants to have a go at being Pac-Man there's genuine incentive to do well. I would even go as far as to say it's one of Miyamoto's greatest games!
Mario Party: The series gets criticized for a) being boring and b) being whored out every year, but in terms of the social aspect I would argue that Mario Party is the ultimate multiplayer game. Because of its gentle pace and lack of intensity you don't have to concentrate particularly hard, which gives you time to chat with your friends. You can sit, chat and hang out with your friends and kind of play Mario Party "in the background". Plus because everything happens on the same screen, everyone is experiencing the same thing so you can really share the experience and interact with each other. Although there is a obviously interaction in something like a Mario Kart race, the split-screen aspect means you are essentially racing your own race.

2 player on Super Mario Bros: I die, you play; you die, I play. I remember one instance where we went five hours straight doing that. Fun times.
Also: Mario Kart, Marvel Vs Capcom, Fighters Megamix, Virtua Cop, Rampage World Tour, Micro Machines, Daytona USA, Halo CE Co-Op, those N64 wrestling games; Pokemon Red, then battling in 3D on Stadium. There's immediately come to mind.

But I think that multiplayer game overall depends on what type of gamer you are. A casual gamer won't care too much if you're playing the latest, top rated game. They prefer just being able to play, an experience which mario kart delivers. On the other hand, I prefer an "intense" match of ssbb. Remember that "fair" quote? "Final destination, no items, 3 stock" All ssb games can be fair, and fun, but you don't here and mario kart players go "rainbow road, no items" because you just can't prove who is better by doing that, and it wouldn't be fun.

Super Mario and Crash Bash hold my fondest memories, I think Crash Bash was the only party game I actually conquered.

I much prefer being in the room. Online kind of bores me...even with voice chat because then I feel kind of...awkward. And I hate being screeched at by little kids calling me obscenities.
As for my favorite games...Goldeneye is still king. Even today when I have people over and we play it (maybe once a year!). Mario Kart 64 and Smash Bros 64 are awesome, too. So is Brawl, of course.
I don't much care for Mario Kart Wii, though. I think it's the frame rate that kills it for me. Everything seems so slick and slidey compared to previous Mario Karts.
Of course, there are also your standby fighters..Marvel vs. Capcom, MKII, Street Fighter II, etc.
I really need to get a second 360 controller someday, too, so I can finally play games like Castle Crashers and TMNT Arcade with someone else.

I've really never liked multiplayer so much because i've never been what you would call "good" at multiplayer. However good i am at the game, i usually get my ass kicked by actual people.
However, my favored experiences are Smash Bros, Strikers, Bomberman, and Mario Kart, online and off

I like both local and online multiplayer. (In fact, online dynasty mode for NCAA Football 09 on the 360 is consuming a lot of my gaming time.)
But I find the Wii lacks local multiplayer variety of a good quality. My family picked up SSBB, and we realized it had become stale, as have games like Mario Party. And several of the action games (which my kids enjoy) are limited to 2 players. I love the Wii, but I'm disappointed in its multiplayer games.
Personally I would LOVE to see a good 4-player action/RPG (local mainly) in the spirit of Baldur's Gate (only 2 player originally) or Champions of Norrath.
The original Everquest, with its severe death penality and difficulty, remains my favorite multiplayer experience.

The best multiplayer game I played in my youth was Street Fighter 2. I basically lived at the arcades on sundays.
Now for some wii games:
RRR (the shooting sections are awesome with co-op)
Wii Sports
Mario Party
Smash Bros
Ghost Squad

Guitar Hero and Mario Kart are the only games I know where any of my friends can pick them up and have fun, and I like that. My friend Katie who effing hates video games will drive with the wheel or drum on the drums. It's awesome there's games that do that. I'd include Wii Sports but I think we all overplayed that one.

Marvel Ultimate Alliance has a great co-op experience with a lot of characters to choose from. Once you get a feel for the controls, it's a blast. Tiger Woods '09 has a great multiplayer experience as well.

I've always had a jolly good time playing EBA against my friend. We would always choose the back up dancers and face off to the same five songs.
Jolly good fun, especially when combined with trying to sing the song while playing.
